Bellicum showcases the latest CAR-T/TCR dual switch technology

On April 4, local time, Bellicum Pharmaceuticals presented two pre-clinical results of CAR-T and TCR drug candidates based on unique dual-switch technology at the American Association for Cancer Research (AACR) 2017 Annual Meeting.

Bellicum developed this dual-switch technology to improve the efficacy, durability and safety of adoptive cell therapies. When the target antigen is present with the rimiducid (small molecule drug), the MyD88/CD40 (iMC) costimulator switch is turned on, and the genetically modified T cells gain proliferation and activation; when a serious toxicity event occurs, rapamycin is injected ( Rapamycin) activates a novel suicide switch called RapaCIDe that triggers rapid apoptosis in cells. This dual-switch technology has been introduced to Bellicum's GoCAR-T and GoTCR development platforms.

In a poster presentation entitled "Double-switched HER2-CAR-T cells: small molecule-controlled GO/STOP switch in vivo targeted therapy for solid tumors," Bellicum scientists tested a HER2-targeting CAR-T cell. Therapy, in which a new dual-switch platform containing iMC costimulatory signals and RapaCIDe apoptotic signals was used. The results show that this new RapaCIDe switch is as effective as the company's CaspaCIDe? switch in inducing apoptosis, and the addition of the iMC costimulator switch enhances T cell proliferation and tumor killing ability. This study demonstrates that Bellicum's dual-switch GoCAR-T technology can effectively control tumor growth and T cell proliferation, activation and clearance in solid tumor models.

In another poster presentation entitled "Double Switching TCR: A Dual Ligand System Controls Proliferation and Apoptosis of PRAME TCR Modified T Cells by Inducing MyD88/CD40 and Caspase-9 Signaling Pathways", Bellicum's Scientists The above two-switch system is also used on the TCR platform. The results demonstrate the multi-platform reproducibility of the technology, and Bellicum's dual-switch TCR technology is the first technical prototype reported to improve the efficacy, persistence, and safety of adoptive T-cell therapies.
